Around the Basin cycle rides spotted the opportunities offered by the $5.4 million trail and signed a commercial lease with the Queenstown Lakes District Council, understood to be costing it about $2000 per year.
Riders can opt for half- or full-day tours, guided or supported by the firm's staff of four.
Owner Steve Norton said the tours would give tourists the benefit of the guides' local knowledge as well as support or a lift when needed.
"It's more of a luxury tour. We will stop by for a coffee at the boat shed in Frankton ... and there will be lifts for those that need them over the harder parts of the trail.
"They'll be well looked after."
Tours will start at the Ride Centre near bottom of the Skyline gondola, meander through Frankton, with a coffee stop, through to Arrowtown and finish at the Gibbston Valley Winery. Riders will be transported back to Queenstown from Gibbston Valley.
The business already has bookings until December 30.
Mr Norton and business partner John Knight would like to see Around the Basin link up with Around the Mountains commercial bike tours eventually, as both trails went hand-in-hand, he said.
"It's the race of the two companies, while both supporting each other. They're sister companies and joined at the hip."
Mr Norton was thrilled with the Queenstown Trail, saying Trails Trust chief executive Kaye Parker had done a top job of pulling it all together.
"She deserves a knighthood.
"She has been the stalwart of this trail."
Mr Norton said now the infrastructure was in place, it was likely more companies would capitalise on Queenstown's newest adventure.
