Civic Events co-promoter Brent Eccles, of Auckland, said in a statement the trio had started rehearsals and promised a two-hour, back-to-back rendition of their hits.
The 2010 tour will involve 18 shows at 16 venues, with 70 people in the tour party.
It will be the first winery tour for Finn and the second for Bic Runga and Dobbyn.
Also in the Winery Tour line-up is Boh Runga, who will perform songs from the Stellar* songbook as well as her solo material.
Boh will be joined on stage by Supergroove frontman and solo performer Che Fu, bringing his own hits such as Fade Away and Misty Frequencies to the set.
Finn said the first Finn-Runga-Dobbyn tour in 2000 was a "proud and passionate moment" for them all.
"We shared ourselves and our songs with audiences from Invercargill to Whangarei, Gisborne to Greymouth, and warmed up the winter with a show that we hope people will never forget.
"This is a summer run in outdoor settings, so we have a chance to revisit without having to re-create. Dave, Bic and I are working on the new show already and are excited by the creative possibilities."
Tickets and bus/ticket packages will go on sale via the Winery Tour website on November 2. Milbrook tickets will also be available via Ticketmaster and Ticket Direct. Tickets will cost between $70 and $85.
