Manawatu led 20-13 at halftime but Otago scored four tries in the second half to win the game.
Star of the show was young winger Jona Nareki, who bagged himself a hat trick of tries.
He was quick and elusive on the wing and outplayed his opposite All Black Nehe Milner-Skudder.
Others to impress for Otago were lock Josh Dickson and halfback Jono Ruru.
Tei Walden also played well in the midfield for Otago as the home team put in some bruising hits on defence.
