Experienced hooker Jason Macdonald will play for Taieri against Pirates at Peter Johnstone Park.
Exciting midfield back Jayden Hayward will play for Zingari-Richmond against Kaikorai at Montecillo.
Halfback Sean Romans will link with his Otago team-mate Chris Noakes in the University A team that plays Alhambra-Union at the North Ground.
"They have not played much Super 14 rugby and do not have any niggling injuries," O'Brien said.
"They have been released by the coaching staff."
O'Brien understood two Southland-based Highlanders would also be released to play club rugby in Invercargill.
