It was the Colours' first win in Dunedin premier rugby for two years and broke a 33 game losing streak.
Dynamic Taieri centre Kieran Moffat scored three of Taieri's five tries when they beat Harbour 25-10 in a repeat of last year's final.
Otago captain Eben Joubert returned to the paddock after a long injury lay off and added steel to the Harbour pack in the second spell.
Dunedin fullback Liam Edwards was in magic touch and scored 34 of Dunedin's points when they beat Green Island 59-23. It included four tries.
Pacy wing Joe Hill scored two of Southern's six tries when they put their foot on the throttle in the second spell to beat Kaikorai 39-15.
Inspirational leadership from Alhambra-Union captain Matt Kissick, who led from the front with his speed to the breakdown and driving play from No 8, helped his side to a 25-19 victory over Pirates.
